dcterms.abstract | 研究了球衣菌 (Sphaerotilussp.)W991 3 6合成聚羟基烷酸 (PHA)的培养基配方及发酵条件。结果表明 ,W991 3 6适宜发酵培养基配方为 :葡萄糖 1 0 2 5g/L ,蛋白胨 2 6 3g/L ,MgSO4·7H2 O 0 1 7g/L ,CaCl2 0 0 5g/L ,NaH2 PO4·2H2 O 0 0 2g/L ,K2 HPO40 0 4g/L ,KH2 PO40 0 3g/L ;最佳接种量为 0 1 3 8g (干 ) / 1 0 0mL ,培养基适宜初始pH为 7 0~ 7 5 ,发酵 3 6h可获得较大的细胞干重与PHA产量 ,经1 H NMR与GC分析 ,其PHA为羟基丁酸与羟基戊酸共聚体P (HB co HV) ,其中羟基戊酸 (HV)含量达 3 9 3 %。 | - |
dcterms.abstract | Fermentation of synthesizing Poly-β-hyd roxyalkanoate(PHA)by Sphaeroti lus sp.W99136 were optimized.The result of uniform design test showed that the o ptimized medium composition contained (g/L):glucose 10 25,peptone 2 63,MgSO 4 ·7H 2O 0 17,CaCl 2 0 05,NaH 2PO 4·2H 2O 0 02,K 2HPO 4 0 04,KH 2PO 4 0 03,The optimum seed c ulture volume was 0 138g(CDW)/100mL.With control initial pH in the medium betwe e n 7 0,higher dry cell weight and PHA content were achieved after fermentation f o r 36h 1H-NMR and GC analysis of PHA products from Sphaero tilus sp.W99136 show that the product was P(HB-co-HV).The HV content of P(HB-c o-HV) came to 39 3%. | - |
