The exopolysaccharide provided by the invention comprises a main chain formed by glucose and mannose, and a side chain formed by connecting galactose with the main chain, wherein the monosaccharide ratio of the mannose to the glucose to the galactose is (3.5 to 4.5) : (6.5 to 7.5) : (0.8 to 1.5) according to the molar ratio; The preparation method of the exopolysaccharide comprises the following steps: performing liquid fermentation on a strain Cs-HK1, and collecting fermentation liquor; carrying out alcohol precipitation treatment and deproteinization treatment on the fermentation liquor to prepare a polysaccharide crude extract; and dissolving the polysaccharide crude extract with water, and carrying out gel column chromatography. The method is simple and convenient, simple to operate and easy for large-scale mass production. Tests show that the exopolysaccharide prepared by the method has good anti-inflammatory activity and shows a good drug application prospect. 本发明属于生物医药技术领域,尤其涉及一种胞外多糖及其制备方法和应用。本发明提供的胞外多糖包括:由葡萄糖和甘露糖形成的主链,由半乳糖连接所述主链形成的侧链;以摩尔比计算,所述甘露糖、葡萄糖和半乳糖的单糖比例为(3.5‑4.5):(6.5‑7.5):(0.8‑1.5)。该胞外多糖的制备方法包括:将菌株Cs‑HK1进行液体发酵,收集发酵液;将发酵液进行醇沉处理以及脱蛋白处理,制得多糖粗提物;将多糖粗提物用水溶解,进行凝胶柱层析。该方法简便,操作简单,易于规模化量产。经测试,由上述方法制得的胞外多糖具有良好抗炎活性,显示出很好的药物应用前景。 |
