Abstract: The invention discloses a formaldehyde detection method of a fluorescence resonance energy transfer probe based on a coupling reaction, which comprises the following steps: adding amine with a fluorophore, alkyne with a fluorophore and a metal catalyst into a reaction medium in which a sample to be detected is dissolved to obtain an original reaction solution; performing fluorescence detection on the original reaction solution, and determining first fluorescence emission peak intensity corresponding to the original reaction solution; heating the original reaction solution, so that all components in the original reaction solution are subjected to a metal catalytic coupling reaction, and obtaining a target reaction solution; performing fluorescence detection on the target reaction solution, and determining second fluorescence emission peak intensity corresponding to the target reaction solution; and determining the formaldehyde concentration in the sample to be detected according to the first fluorescence emission peak intensity and the second fluorescence emission peak intensity. Fluorescence detection is adopted and matched with fluorescence resonance energy transfer, interference of background signals is reduced, the elution step is omitted, detection is convenient and fast, the cost is low, and formaldehyde selectivity is high.
本发明公开了一种基于偶联反应的荧光共振能量转移探针的甲醛检测方法,包括:在溶解有待测样品的所述反应介质中加入带荧光基团的胺类、带荧光基团的炔烃以及金属催化剂,得到原始反应溶液;对原始反应溶液进行荧光检测,确定原始反应溶液对应的第一荧光发射峰强度;对原始反应溶液进行加热,使原始反应溶液中的各组分发生金属催化偶联反应,得到目标反应溶液;对目标反应溶液进行荧光检测,确定目标反应溶液对应的第二荧光发射峰强度;根据第一荧光发射峰强度和第二荧光发射峰强度,确定待测样品中的甲醛浓度。本发明采用荧光检测,配合荧光共振能量转移降低背景信号的干扰,省去洗脱步骤,使检测便捷,成本廉价且对甲醛选择性高。
