Abstract: The invention provides small-interference RNA for inhibiting human hephaestin (HEPH) gene expression. The small-interference RNA is reversely complemented with human HEPH mRNA, and the length of the small-interference RNA is 19 to 27bp. The invention further discloses a DNA sequence for coding the corresponding shRNA of the small-interference RNA and recombinant slow virus capable of expressing the shRNA. The small-interference RNA, the DNA for coding the shRNA and the recombinant slow virus can effectively inhibit in vivo human HEPH expression, inhibit the release and metabolism of iron and adjust the serum iron level in vivo so as to fulfill the purpose of treating iron metabolism related diseases.
本发明提供了一种抑制人膜铁转运蛋白辅助蛋白基因(HEPH)表达的小干扰性RNA,所述小干扰性RNA与人膜铁转运蛋白辅助蛋白基因mRNA反向互补,长度为19-27bp。本发明进一步公开了编码所述小干扰性RNA的相应shRNA的DNA序列以及能表达所述shRNA的重组慢病毒。本发明的小干扰性RNA、编码shRNA的DNA以及重组慢病毒能够有效抑制体内人膜铁转运蛋白辅助蛋白表达、抑制铁的释放与代谢,调节体内的血清铁水平,从而达到治疗铁代谢相关疾病的目的。
